Will be in St. John for a week and St Thomas for a week. We'd like to spend a day in Tortola exploring as well. Any suggestions on round trip transportation from either US islands for such a day trip?

Take the ferry -- it is easy and quick. There are two drop-off spots for the ferries, Road Town (the main town) and West End.
There are several ferries that run daily from both STT and STJ. I believe the latest ferry back to STT from Road Town leaves at around 5:30 P.M.
You will find the ferry schedules listed in many of the free tourist publications that you will find all over on both islands and at the airport. It is always a good idea to call on the morning of your departure to confirm the schedule.
A good magazine to get your hands on is "St. Thomas This Week." It's free and widely available. Below is also link to their website:

You might want to consider renting a car once you get to Tortola. If you stop at the visitors center in Road Town after you arrive, they can arrange everything for you, but you might also want to consider making a reservation in advance. If the rental company you use is not within walking distance from the dock, most companies will come pick you up at the dock and then take you back when you return the car.

There is a ferry from STJ to Tortola's West End at 8:30AM. Week days the return ferry is at 4:15PM (5:30 on Fridays). Rent a car there and off you go.
